Bio-Activity | Measured by the ability of the immobilized protein to support the adhesion of NIH_x001e_3T3 mouse embryonic fibroblast cells. When 5 x 104 cells/well are added to recombinant mouse Nectin-4 coated plates (10 μg/mL with 100 μL/well), approximately 30%-40% will adhere after 1 hour incubation at 37 °C.

Background
Nectin-4 (PVRL4) is a type I transmembrane glycoprotein which belongs to the nectin family of Ig superfamilyproteins. It contains two Ig-like C2-type domains and one Ig-like V-type domain. PVRL4 seems to be involved incell adhesion through trans-homophilic and -heterophilic interactions, the latter including specificallyinteractions with nectin-1. It does not act as receptor for alpha-herpesvirus entry into cells. It is predominantlyexpressed in placenta, the embryo and breast carcinoma. But it is not detected in normal breast epithelium.The soluble form is produced by proteolytic cleavage at the cell surface (shedding), probably by ADAM17.Mutations in this gene are the cause of ectodermal dysplasia-syndactyly syndrome type 1, an autosomalrecessive disorder.

Usage guide
Storage | Store at -20℃.Store the lyophilized protein at -20℃ to -80 ℃ up to 1 year from the date of receipt. After reconstitution, the protein solution is stable at -20℃ for 3 months, at 2-8℃ for up to 1 week. |

Reconstitution | Centrifuge the vial before opening. Reconstitute to a concentration of 0.1-0.5 mg/mL in sterile distilled water. Avoid vortex or vigorously pipetting the protein. For long term storage, it is recommended to add a carrier protein or stablizer (e.g. 0.1% BSA, 5% HSA, 10% FBS or 5% Trehalose), and aliquot the reconstituted protein solution to minimize free-thaw cycles. |
