Description
Lb Cas12a Nuclease from Lachnospiraceae bacterium is a site-specific DNA endonuclease guided by a single 40-44 nucleotide guide RNA (crRNA). Targeting requires a crRNA complementary to the target site as well as a 5’ TTTV protospacer adjacent motif (PAM) on the DNA strand opposite the target sequence. Cleavage by LbCas12a Nuclease occurs ~17 bases 3′ of the PAM and leaves overhanging ends.This product also has a trans-cleavage activity, which produces high non-specific ssDNA cleavage activity when the target dsDNA, crRNA, and Cas12a protein form a ternary complex.

1. Cis-cleavage activities
LbCas12a specifically recognizes and cleaves the pUC19 sequence under the guidance of crRNA, causing double stranded breaks in the target DNA, cutting the substrate with a total length of about 2600bp into two fragments of 1750bp and 850bp.
2. Trans-cleavage activities
Collect fluorescence signals using a fluorescence quantitative PCR instrument, and collect fluorescence signals every 60 seconds. LbCas12a forms a ternary complex with a specific target dsDNA under the guidance of crRNA, it will be activated for efficient trans cleavage activity against ssDNA, and the ssDNA reporter probe in the system will be fragmented to emit a fluorescence signal.
