Western blot analysis of lysates from wild type (WT) and 293T cells transfected with CD8β, using CD8β Rabbit mAb (A24827) at 1:80000 dilution.
Secondary antibody: HRP-conjugated Goat anti-Rabbit IgG (H+L) (AS014) at 1:10000 dilution.
Lysates/proteins: 25μg per lane.
Blocking buffer: 3% nonfat dry milk in TBST.
Detection: ECL Basic Kit (RM00020).
Exposure time: 20s.

Confocal imaging of paraffin-embedded human tonsil using CD8β Rabbit mAb (A24827, dilution 1:200) followed by a further incubation with Cy3 Goat Anti-Rabbit IgG (H+L) (AS007, dilution 1:500) (Red). DAPI was used for nuclear staining (blue). Objective: 40x.

Confocal imaging of Jurkat cells using CD8β Rabbit mAb (A24827, dilution 1:200) followed by a further incubation with Cy3 Goat Anti-Rabbit IgG (H+L) (AS007, dilution 1:500) (Red). DAPI was used for nuclear staining (blue). Objective: 100x.

Flow cytometry: 1X10^6 293F cells (negative control, left) and 293F (Transfection, right) cells were surface-stained with CD8β Rabbit mAb (A24827, 2 μg/mL, orange line) or ABflo® 647 Rabbit IgG isotype control (A22070, 5 μl/Test, blue line), followed by Alexa Fluor® 647 conjugated goat anti-rabbit pAb staining. Non-fluorescently stained cells were used as blank control (red line).

Flow cytometry: 1X10^6 293F (Transfection) cells were surface-stained with ABflo® 647 Rabbit IgG isotype control (A22070, 5 μl/Test, left) or CD8β Rabbit mAb (A24827, 2 μg/mL, right).

Flow cytometry: 1X10^6 Human PBMC were surface-stained with CD8β Rabbit mAb (A24827, 2 μg/mL, orange line) or ABflo® 647 Rabbit IgG isotype control (A22070, 5 μl/Test, blue line). Non-fluorescently stained Human PBMC were used as blank control (red line).

Flow cytometry: 1X10^6 Human PBMC were surface-stained with ABflo® 647 Rabbit IgG isotype control (A22070, 5 μl/Test, left) or CD8β Rabbit mAb (A24827, 2 μg/mL, right).

Background

The CD8 antigen is a cell surface glycoprotein found on most cytotoxic T lymphocytes that mediates efficient cell-cell interactions within the immune system. The CD8 antigen, acting as a coreceptor, and the T-cell receptor on the T lymphocyte recognize antigens displayed by an antigen presenting cell (APC) in the context of class I MHC molecules. The functional coreceptor is either a homodimer composed of two alpha chains, or a heterodimer composed of one alpha and one beta chain. Both alpha and beta chains share significant homology to immunoglobulin variable light chains. This gene encodes the CD8 beta chain isoforms. Multiple alternatively spliced transcript variants encoding distinct membrane associated or secreted isoforms have been described. A pseudogene, also located on chromosome 2, has been identified.
