Western blot analysis of various lysates using CD74 Rabbit mAb (A24027) at 1:3000 dilution.
Secondary antibody: HRP-conjugated Goat anti-Rabbit IgG (H+L) (AS014) at 1:10000 dilution.
Lysates/proteins: 25μg per lane.
Blocking buffer: 3% nonfat dry milk in TBST.
Detection: ECL Basic Kit (RM00020).
Negative control (NC):Jurkat
Exposure time: 10s.
Immunohistochemistry analysis of paraffin-embedded Human tonsil tissue using CD74 Rabbit mAb (A24027) at a dilution of 1:100 (40x lens). High pressure antigen retrieval performed with 0.01M Tris/EDTA Buffer (pH 9.0) prior to IHC staining.
Immunofluorescence analysis of Raji cells using CD74 Rabbit mAb (A24027) at a dilution of 1:100 (40x lens). Secondary antibody: Cy3-conjugated Goat anti-Rabbit IgG (H+L) (AS007) at 1:500 dilution. Blue: DAPI for nuclear staining.
Flow cytometry:1X10^6 Jurkat cells (negative control, left) and Raji cells (right) were surface-stained with CD74 Rabbit mAb (A24027, 2 μg/mL, orange line) or PE Rabbit IgG isotype control (5 μl/Test, blue line), followed by PE Donkey anti-rabbit Antibody staining. Non-fluorescently stained cells were used as blank control (red line).
