Western blot analysis of lysates from rat brain, using Phospho-Tau-S404 Rabbit mAb (AP1378) at 1:5000 dilution. Rat brain cells were treated by λ-PP mixed solution (1ul) at 30℃ for 30 minutes.
Secondary antibody: HRP-conjugated Goat anti-Rabbit IgG (H+L) (AS014) at 1:10000 dilution.
Lysates/proteins: 25μg per lane.
Blocking buffer: 3% nonfat dry milk in TBST.
Detection: ECL Basic Kit (RM00020).
Exposure time: 10s.

Western blot analysis of lysates from Mouse brain using Phospho-Tau-S404 Rabbit mAb (AP1378) at 1:5000 dilution. Mouse brain lysates were treated by λ-PP mixed solution (1ul) at 30℃ for 30 minutes.
Secondary antibody: HRP-conjugated Goat anti-Rabbit IgG (H+L) (AS014) at 1:10000 dilution.
Lysates/proteins: 30 μg per lane.
Blocking buffer: 3% nonfat dry milk in TBST.

Western blot analysis of lysates from SH-SY5Y cells using Phospho-Tau-S404 Rabbit mAb (AP1378) at 1:5000 dilution. SH-SY5Y cells were treated by Okadaic Acid (100 nM) at 37℃ for 1 hour and Calyculin A (100nM) at 37℃ for 30 minutes after serum-starvation overnight.
Secondary antibody: HRP-conjugated Goat anti-Rabbit IgG (H+L) (AS014) at 1:10000 dilution.
Lysates/proteins: 30 μg per lane.
Blocking buffer: 3% nonfat dry milk in TBST.
Detection: ECL Basic Kit (RM00020).

Background

This gene encodes the microtubule-associated protein tau (MAPT) whose transcript undergoes complex, regulated alternative splicing, giving rise to several mRNA species. MAPT transcripts are differentially expressed in the nervous system, depending on stage of neuronal maturation and neuron type. MAPT gene mutations have been associated with several neurodegenerative disorders such as Alzheimer's disease, Pick's disease, frontotemporal dementia, cortico-basal degeneration and progressive supranuclear palsy.
