Western blot analysis of various lysates, using Phospho-Tau-T181 antibody (AP1387) at 1:2000 dilution.Mouse brain were treated by λ-PP mixed solution (1ul) at 30℃ for 30 minutes.
Secondary antibody: HRP Goat Anti-Rabbit IgG (H+L) (AS014) at 1:10000 dilution.
Lysates/proteins: 25μg per lane.
Blocking buffer: 3% nonfat dry milk in TBST.
Detection: ECL Enhanced Kit (RM00021).
Exposure time: 60s.

Immunohistochemistry analysis of Phospho-Tau-T181 in (5XFADJ)Mouse brain (left) and normal mouse brain (right) using Phospho-Tau-T181 Rabbit mAb(AP1387) at a dilution of 1:200 (40x lens). High pressure antigen retrieval was performed with 0.01 M citrate buffer (pH 6.0) prior to IHC staining.

DAPI was used for nuclear staining (Blue). Objective: 40x.Perform microwave antigen retrieval with 10 mM citrate buffer pH 6.0 before commencing with IF staining protocol.

Background

This gene encodes the microtubule-associated protein tau (MAPT) whose transcript undergoes complex, regulated alternative splicing, giving rise to several mRNA species. MAPT transcripts are differentially expressed in the nervous system, depending on stage of neuronal maturation and neuron type. MAPT gene mutations have been associated with several neurodegenerative disorders such as Alzheimer's disease, Pick's disease, frontotemporal dementia, cortico-basal degeneration and progressive supranuclear palsy.
