Western blot analysis of lysates from from 293T cells transfected with CDK5+CDK5R1+Tau and CDK5+CDK5R1+mutant Tau(T181/T217A) using Phospho-Tau-T217 Rabbit mAb (AP1373) at 1:10000 dilution.
Secondary antibody: HRP Goat Anti-Rabbit IgG (H+L) (AS014) at 1:10000 dilution.
Lysates/proteins: 25μg per lane.
Blocking buffer: 3% nonfat dry milk in TBST.
Detection: ECL Enhanced Kit (RM00021).
Exposure time: 60s.

Confocal imaging of paraffin-embedded 5xFAD mouse brain and mouse brain using Phospho-Tau-T217 Rabbit mAb (AP1373, dilution 1:200) followed by a further incubation with Cy3 Goat Anti-Rabbit IgG (H+L) (AS007, dilution 1:500)(Red).DAPI was used for nuclear staining (Blue). Objective: 40x.Perform microwave antigen retrieval with 0.01 M citrate buffer (pH 6.0) prior to IF staining.

Background

This gene encodes the microtubule-associated protein tau (MAPT) whose transcript undergoes complex, regulated alternative splicing, giving rise to several mRNA species. MAPT transcripts are differentially expressed in the nervous system, depending on stage of neuronal maturation and neuron type. MAPT gene mutations have been associated with several neurodegenerative disorders such as Alzheimer's disease, Pick's disease, frontotemporal dementia, cortico-basal degeneration and progressive supranuclear palsy.
