Western blot analysis of lysates from wild type (WT) and p53 knockout (KO) 293T cells using [KO Validated] p53 Rabbit PolymAb® (A26037PM) at 1:3000 dilution incubated overnight at 4℃.
Secondary antibody: HRP-conjugated Goat anti-Rabbit IgG (H+L) (AS014) at 1:10000 dilution.
Lysates/proteins: 25 μg per lane.
Blocking buffer: 3% nonfat dry milk in TBST.
Detection: ECL Basic Kit (RM00020).
Exposure time: 15s.

ABclonal:Immunofluorescence - [KO Validated] p53 Rabbit PolymAb® (A26037PM)

Confocal imaging of A-431 cells using [KO Validated] p53 Rabbit PolymAb® (A26037PM, dilution 1:200) followed by a further incubation with Cy3 Goat Anti-Rabbit IgG (H+L) (AS007, dilution 1:500) (Red). The cells were counterstained with α-Tubulin Mouse mAb (AC012, dilution 1:400) followed by incubation with ABflo® 488-conjugated Goat Anti-Mouse IgG (H+L) Ab (AS076, dilution 1:500) (Green). DAPI was used for nuclear staining (Blue). Objective: 100x.

Immunohistochemistry analysis of paraffin-embedded Human lung squamous carcinoma tissue using [KO Validated] p53 Rabbit PolymAb® (A26037PM) at a dilution of 1:200 (40x lens). High pressure antigen retrieval performed with 0.01M Tris-EDTA Buffer(pH 9.0) prior to IHC staining.

Background

This gene encodes a tumor suppressor protein containing transcriptional activation, DNA binding, and oligomerization domains. The encoded protein responds to diverse cellular stresses to regulate expression of target genes, thereby inducing cell cycle arrest, apoptosis, senescence, DNA repair, or changes in metabolism. Mutations in this gene are associated with a variety of human cancers, including hereditary cancers such as Li-Fraumeni syndrome. Alternative splicing of this gene and the use of alternate promoters result in multiple transcript variants and isoforms. Additional isoforms have also been shown to result from the use of alternate translation initiation codons from identical transcript variants (PMIDs: 12032546, 20937277).
